Description

These PDPN-Cre transgenic mice express cre recombinase in Pdpn-expressing lymphatic endothelial cells (LECs). When these mice are bred with mice containing a loxP-flanked sequence of interest, Cre-mediated recombination will result in deletion of the floxed sequences in Pdpn-expressing LECs of the offspring. These mice may be useful for studies related to the role of Pdpn-expressing cells in cancer progression and tumor metastases.

Of note, the donating investigator has also made this transgene available on a congenic C57BL/6J background as Stock No. 039769.

Detailed description

These PDPN-Cre transgenic mice express cre recombinase under control of the mouse Pdpn (Podoplanin) gene promoter. When these transgenic mice are bred with mice containing loxP-flanked sequences, Cre-recombinase activity is detected in Pdpn-expressing lymphatic endothelial cells (LECs) of the offspring. The donating laboratory bred these PDPN-Cre transgenic mice to mice expressing a Cre-inducible (LSL)-yellow fluorescent protein (YFP) on a BALB/c background and reports that YFP expression was detectable in PDPN-positive lymphatic endothelium in healthy tissues from the resulting progeny. LECs isolated from IFA-induced lymphangioma in these PDPN-Cre;LSL-YFP double transgenic mice also demonstrated significant YFP expression (80% of all nucleated cells) within the lymphangioma tissue. Hemizygous PDPN-Cre mice are viable and fertile. Homozygous viability/fertility have not been tested by the donating laboratory. This strain may be useful for understanding the role of PDPN positive LECs in tumor progression and metastases.

Development

The PDPN-Cre transgenic construct was generated by cloning and ligating the 4kb mouse Podoplanin (Pdpn) promoter sequence to a full length cre recombinase sequence and an SV40 polyA signal using the pCR2.1-TOPO vector (Invitrogen). The construct was microinjected into FVB fertilized mouse eggs and founder line H was subsequently established. The donating investigator confirmed that a single copy of the PDPN-TK transgene integrated on chromosome 16. The mice were backcrossed to BALB/cAnNCrl for more than twenty generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize BALB/cByJ oocytes (Stock No. 001026).

Of note, the donating laboratory indicates that the Y chromosome was fixed during backcross to BALB/c.
